Extracting fruits and veggie DNA using Biorad DNA buffer

Post by candy3105 »

Hello, I selected a project for Science Fair at my school to test if the Cheek cells and Strawberry have DNAs using a Biorad "Genes in a bottle kit". But my ISD has not allowed me to use cheek cells, so now I'm concentrating on testing DNA in different fruits and veggies. Has anyone done this experiment to test fruits other than strawberries and vegetables? I'm not sure if it is feasible. I have ran out of the little DNA buffer included with the Biorad kit and now to do more tests I will have to buy and spend more or create some buffer myself. Not sure how. Will the same kind of buffer(dentaured alcohol, as mentioned in the kit guide) be suitable to test veggies? I have so many questions. Please help.

Hey candy3105,

You can totally still complete this DNA Extraction experiment without using the Bio-Rad kit!
Recipe to make your own DNA Extraction buffer: https://www.biologycorner.com/worksheet ... ction.html
A mixture of detergent (which lyses, or bursts open, cells), salt (which causes the DNA to precipitate from the lysed cell), and water serve as good DNA extraction buffer!
If you need help conducting the strawberry DNA extraction, this is another helpful website: https://www.scientificamerican.com/arti ... awberries/

If you want to do another DNA extraction lab that doesn't involve strawberries, Science Buddies has great projects like this one: https://www.sciencebuddies.org/science- ... p001.shtml

Hope this was helpful,

Hello JSK,
Your advice was very helpful. I was able to make the DNA extraction buffer at home and tested a fruit today. Will try testing with more fruits/veggies also.
You were so helpful. Thank you very much!
