lactobacillus acidophilus science experiment help

I've been trying to post this question online but can't find the link/box to post a question.
Anyhow, I want to do a science fair experiment on the "Effect of papaya seeds extract on lactobacillus acidophilus and E. Coli "
Do you know what would be the best way to test this? I know it is easy to measure inhibition area for E.Coli on agar petri plates with filter disk in the center. However, I read on your site and some other sites that lactobacillus acidophilus does not survive well on agar petri dish. The survive better in broth. What kind of materials, broth, tubes, agar plates etc... should I get?
What materials do I need to do this experiment?
I want to measure the growth rate of lactobacillus acidophilus with different concentrations ofpapaya seed extract. How can I measure the inhibition area or count on lactobacillus acidophilus"?
Also, what in innoculation? How is this done? How do you remove a hole in agar plate to fill it with testing items? Does this also test inhibition area?
