How could you demonstrate that it is a structure on the bottom side of leaves that regulates the amount of transpiration that occurs in a plant?

You have an excellent question.

Here is a simple way to measure the amount of water lost by a plant: You simply trap water emitted from plants in plastic bags and weigh the bags before and after transpiration to measure the water.

Are you doing a science project on plant transpiration? You could also measure the stomatal density using a microscope.
