Science Buddies: "Ask an Expert"

Hi all,
I was wondering if you know of where I could find information on specific strains of E. coli. Specifically, I am going to be working with E. coli K12 strain dg98. I know it is considered biosafety level 1, but... my teacher says that I need more information on the strain to write in my research proposal. (Is it even necessary to write about the specific strain of bacteria? If so, what is the significance; how will it affect my experiment? I'm only using the bacteria to amplify my plasmids.)

Thank you, but the links you posted seem like... a google search or something of that sort. I have looked at google searches, and I got a similar result. The web sites are not very helpful in providing a background of this particular strain, so that was why I asked on this forum. Does anybody know of any books, even, that could possibly help me on this?

Hi M,

Certain strains of E.coli have been engineered to be good for slightly different tasks such amplifying plasmids or producing recombinant proteins.

If you are only using this strain to amplify up plasmids then it is probably because its a high copy number strain - ie. will have many copies of plasmid per bacterial cell so the yield on your plasmid prep will be high. It may also have been engineered to make sure that the plasmids are replicated with as few errors as possible. Some strains are good for plasmids that have a particular backbone, if the original version of the plasmid (and its origin of replication) came from a particular strain it will grow well in that strain.

If you are buying the cells from a supplier it will have a blurb about these features in the catalogue - so i would check the catalogues from companies like Stratagene, NEB, Invitrogen. Also check the details of your plasmid - if its based on a commerical plasmid like pGEM the notes that came with it may recommend a particular strain and say why.

Check with your mentor on this one as to why they use this strain. If you can't get hold of them try a student or postdoc in their lab as they should also know.
