I read the procedure for growing yeast colonies and shining UV rays on them. Should I be growing BOTH the UV sensitive yeast cultures and the wild-type yeast cultures simultaneously using the same procedures?

Yes, you should use the same procedures and time frame for both the UV-sensitive yeast cultures and the wild-type yeast cultures. This way, you have a control strain to compare the UV sensitivity of the genetically engineered strain.

Thus, you would be comparing the effects of BOTH the dilution and the gene knockout on the UV sensitivity of the yeast.
