dose depth affect bacteria growth in any way such as what kinds of bacteria grow at different depths or amounts of bacteria

i have 6 questions that dont all need to bee answerd at once but it would bee very use-full
1. what is the best way to test bacteria amounts on two diferent objects (wood,metal)?
2. what kinds of materials would bee needed for said experiment?
3.how long would it take?
4.would the bactiria from the experiment be dangerous?
5. where would it need to take place?
6. would it need to take place in water or dry land?

That's certainly interesting and important. I don't know anything personally about this topic myself, but here's how I'd approach it.
You can get some good orientation for starting your research on ScienceBuddies. For example, this page discusses sources of information:

You could look for library books about bacteria, water supplies, or health (water contamination).
I tried searching also on Google for "bacteria in water streams." There I learned that this question relates to health issues, and many of the references focus on water supplies.
You asked more specifically about growth on pine wood and steel. Try searching also for "bacteria on wood" -- it appears again that researchers focus on health (are wood cutting boards safer than plastic boards?). Also try "bacteria on metal surfaces" -- you'll find many interesting links.
