Hi,

This is a unique and very interesting topic. There is a commercially available elisa assay for potato virus available at this website. You can download information about the assay, or send an e-mail inquiry to the company asking for additional information. You would need a microplate reader with a 405 nm filter to read this assay:

Here is a protocol for developing an elisa assay from another company that sells the reagents for doing this. You would need an antibody to the potato virus and another antibody to the anti-potato virus antibody to develop your own assay. Notice the extensive troubleshooting guide to help solve the problems that are commonly encountered when developing your own assay.

I am attaching an article describing a polymerase chain reaction method for detecting potato virus. This would be a good option if you had access to a PCR instrument.
