Human DNA Extraction buffer

Post by hknicholas »

Dear Expert,

I am writing to ask for your help on designing a human cheek cell dna extraction buffer. I found many recipes on the web but most of them consist of harmful chemicals such as Tris, SDS etc.. I'd like to extract the dna and run it on a gel although it is not clear. I want to ask you a few questions:
1. Can you provide a recipe for the buffer that the effect of it is similar to those once on the web with harmless chemicals?
2. Can you tell me the amount of the buffer, NaCl solution and alcohol I have to add?
3. What preserve should I do before I load the sample into the 'well' on the gel?

It's a bit urgent, please reply as soon as possible, Thanks!

Hi hknicholas,

Have you tried:
http://www.umbi.umd.edu/~scitech/pdf/DNA.pdf ?
Please see the text below: "Releasing the DNA…" on page 4.

The chemicals described there: detergents, soaps and contact lens solution do not appear to be harmful.

Please excuse if you've already seen this page. Thanks.
Cheers!

Thanks David,

I would like to ask if tris base and edta is a must to dna? What are there purpose in extraction?
Also the dna extracted can we run through it with a agarose gel?
