It sounds like you are working on an interesting project.

Here is a website that shows how to collect fungal samples form wood.

And, here is a website that includes good photographs showing the technique for inoculating fungal samples onto an agar plate. You would need to use a straw or similar device that could hold a small sample of the wood you are sampling.
