A couple questions...
How would you calculate the release of iron?

Can you please explain a bit more about the question you are trying to test for your experiment? Your hypothesis would also be very helpful to know. If you don't have a question or hypothesis yet, Science Buddies has a couple of great articles to help you develop them:

I can answer a couple of your questions now, but it will be much harder to answer the others without some additional information about your experiment.
RE: Is vinegar a good acid to simulate stomach acid?
Stomach acid is mostly hyrdochloric acid (HCl) while vinegar is acetic acid (CH3COOH). You could certainly create an acetic acid solution with the same pH (and therefore the same hydrogen ion concentration) as stomach acid. But, whether that is a suitable model for the stomach depends on what question you are trying to answer.
RE: How does pill breakdown work?
The stomach's juices dissolve pills. Different types of pills have different coatings that can speed up or slow down the dissolution process. This Wikipedia article explains the basic process of dissolution: http://en.wikipedia.org/wiki/Dissolution_(chemistry).
