Methods of methylation of rat liver enzyme

I am Phd student . i am working on rat liver and brain enzymes.
I want to isolate the enzyme from rat liver and brain which converts sphingosine into dimethylsphingosine.
How to extarct the enzyme from rat liver or brain using lysate buffer?
What should be the pH of lysate buffer i should keep to isolate that enzyme. Also i need methylation procedures for it.
Kindly let me know the same.

Hi Sacksant,
This forum is devoted to high school students and younger who are working on projects for science fairs, so your questions are more advanced and specific that the ones that usually come up in this forum.

Myself, I am not familiar with this area of research. I did a quick search on Web of Science to see if I could spot any appropriate journal articles to assist you, but I did not find any. Perhaps some other forum members here may be able to help.
