Hello!
I am stuck on a step for my current science project, where I will need to observe the microscopic properties of tomato tissues after exposing them to allelopathic VOCs. Does anyone know/have any advice on if there is a standard procedure for fixation, dehydration, and preparation of tomato tissues, specifically from the leaves, stem and roots? I am hoping to embed them in paraffin wax and use a microtome.
I would really appreciate any help! Thanks!

Re: Fixation/Preparation of Plant Tissues for Microscope Observation
Hi,
A good starting point for plant histology is FAA (formalin-acetic acid-alcohol) fixation, which preserves plant tissue structure for microscopic viewing.
Here are some useful links to explore more about the topic and methods:
- Overview of FAA: https://plantscience.psu.edu/research/l ... ing-tissue
- FAA solution safety data sheet: https://www.fishersci.com/store/msds?pa ... anguage=en
You can also ask a science teacher or professional for guidance on the procedures and safety protocols depending on your specific experiment.
Hope your project goes well!
