HELP RNAi SCI FAIR PROJECT - How to make a mutant to silence a gene

Post by deleted-511736 »

I am trying to silence a C. Elegan gene but I am not sure where to find or make the mutants.. I don't know how because the general instructions online aren't descriptive. The science buddies kit online is for an entire class and not for an individual student, I am not sure where to start but I have done so much research.. I need someones help.
PLEASE respond !
Thank you,

Hi Angie. I have molecular biology experience and will try to help you. I did a quick search for RNA interference in C elegans and found several papers describing how to use double-stranded RNA (dsRNA) to silence a gene by blocking translation of complimentary mRNAs. Here's one example: http://labs.bio.unc.edu/Goldstein/Dudle ... nMMB05.pdf

Is that the method you were planning to use for RNAi?

Which gene are you trying to silence? Have you worked with C elegans before? Are you experienced in using equipment and reagents in a research lab setting? Will you be able to do your experiments in a well-equipped lab? I need to know where you will be working so I will know what types of experiment you will be able to perform and what techniques you will be able to use.

Please post again with more information and I will try to answer your questions and get you on the right track.
